    功能This gene encodes the D4 subtype of the dopamine receptor. The D4 subtype is a G-protein coupled receptor which inhibits adenylyl cyclase. It is a target for drugs which treat schizophrenia and Parkinson disease. Mutations in this gene have been associated with various behavioral phenotypes, including autonomic nervous system dysfunction, attention deficit/hyperactivity disorder, and the personality trait of novelty seeking. This gene contains a polymorphic number (2-10 copies) of tandem 48 nt repeats; the sequence shown contains four repeats.DRD4 expression has been reported in various regions of the brain as well as in adrenal gland, artery, eye, heart, kidney, placenta, spinal cord, testis, and vas deferens.

    背景资料Members of the G protein-coupled receptor family are distinguished by their slow transmitting response to ligand binding. These seven- transmembrane proteins include the adrenergic, serotonin and dopamine receptors. The effect of the signaling molecule can be excitatory or inhibitory depending on the type of receptor to which it binds. b-adrenergic receptor binds to adrenaline and activates adenylyl cyclase, while a2-adrenergic receptor binds to adrenaline and inhibits adenylyl cyclase. The dopamine receptors are divided into two classes, D1 and D2, which differ in their functional characteristics in that D1 receptors stimulate adenylyl cyclase, while D2 receptors inhibit adenylyl cyclase activity. Five different subtypes of dopamine receptor have been described to date. D1DR and D5DR belong to the D1 subclass, while D2DR, D3DR and D4DR belong to the D2 subclass of dopamine receptors.
