| 背景资料 | Platelet-derived growth factor is a potent mitogen for cells of mesenchymal origin. Binding of this growth factor to its affinity receptor elicits a variety of cellular responses. It is released by platelets upon wounding and plays an important role in stimulating adjacent cells to grow and thereby heal the wound. [SUBUNIT] Antiparallel disulfide-linked dimer of nonidentical (A and B) chains. Homodimers of A and B chains are implicated in transformation processes. A-A and B-B, as well as A-B, dimers can bind to the PDGF receptor. Belongs to the PDGF/VEGF growth factor family. |

Protocol
The cells were fixed with 2% Paraformaldehyde (10 min) , then permeabilized with 90% ice-cold methanol for 30 min on ice.Cells stained with Primary Antibody for 30 min at room temperature. The cells were then incubated in 1 X PBS/2%BSA/10% goat serum to block non-specific protein-protein interactions followed by the antibody for 15 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
Primary Antibody (green line): Rabbit Anti-PDGF-B antibody (bs-0185R)
Dilution: 1μg /10^6 cells;
Isotype Control Antibody (orange line): Rabbit IgG .
Secondary Antibody (white blue line): F(ab’)2 fragment goat anti-rabbit IgG-FITC
