Lucigen's CopyRight® v2.0 BAC cloning kits with pSMART® BAC (Figure1) or pEZ BAC (Figure 2) vector and BAC-Optimized Replicator™ v2.0 Electrocompetent Cells result in unprecedented accuracy, reliability, and efficiency in BAC cloning and library construction. These vectors incorporate the CloneSmart® transcription-free technology, which minimizes cloning bias and enables cloning of otherwise toxic genes, along with inducible copy number amplification (20-50X) for high DNA yields and a lacZ-sacB stuffer fragment that eliminates uncut vector from recombinants, resulting in zero background. Unlike other BAC cloning systems, Lucigen's CloneSmart technology assures that even "unclonable" inserts are stably maintained. Sequences are not lost or rearranged in the cloning process. CopyRight Kits offer superior performance for BAC cloning at a much better price than other kits. Figure 3 shows an example of aRandom Shear BAC library generated with pSMART BAC.

Each CopyRight® v2.0 BAC Cloning Kit contains: pre-cut vector (pSMART BAC with a choice of BamHI or HindIII sites), CopyRight DNA Ligase, CopyRight 5X Ligation Buffer, Positive Control Insert DNA, Sequencing Primers, BAC-Optimized Replicator v2.0 Electrocompetent Cells in DUO packaging (2 transformations per tube), Positive Control Insert DNA, Recovery Medium, Arabinose Induction Solution, and complete protocols. Each CopyRight pEZ BAC Cloning Kit contains: pre-cut CopyRight vector (pEZ BAC Blunt), CloneSmart DNA Ligase, CloneDirect™ 10X Ligation Buffer (includes ATP), Positive Control Insert DNA, Replicator Induction Solution, Sequencing Primers, and complete protocols. Kits with cells also include BAC-Optimized Replicator v2.0 Electrocompetent Cells (>1 × 1010 cfu/µg DNA) packaged in two transformations per tube (DUOs), Positive Control Plasmid, and Recovery Medium.
Please note that the following vectors have been discontinued: pSMART BAC EcoRI (catalog numbers 42031-1, 42031-2) and pEZ BAC BamHI (catalog numbers 42007-1, 42007-2 and 42012-1).”
