文献和实验,38]是将人胰腺癌1990细胞以2×105个/ml种入96孔细胞培养板，37℃5%CO2培养箱培养4-6小时，用完全培养液（加有1.2µg/m的放线菌素D）做梯度稀释的样品加入细胞板（n=3）。置37℃5%CO2培养箱，18h后弃上清，以结晶紫固定液（结晶紫0.5%，甲醛8%，NaCl0.1%，乙醇20%）染色15min，蒸馏水洗去结晶紫。每孔再加入200μ33%的乙酸，旋涡混匀，置酶联仪读出D（595）值，以未加细胞的空白孔为D本底。根据活性单位定义：使培养孔中的细胞50%死亡为一个单位

蛋白提取：所在操作冰上进行 （1）取组织，用PBS冲洗干净组织上的血液. 加入 10ml Buffer A , 用剪刀尽可能剪碎组织,于冰上充分匀浆。每次30秒,重复3-5次. （2）离心机 1000rpm，4℃ 离心 10min 后，所得上清液转入超速离心管。(去掉大块组织及结缔组织) （3）100000g，4℃ 离心 1hr, 弃去上清.(此为胞浆蛋白,若只提取胞浆蛋白,可用10000rpm, 4度, 30分钟离心，取上清即可 )。沉淀用适量的 Buffer B重悬，4度过夜后，分装至 EP
