这是在网上看到的，觉得还行，大家看一下1、载体和插入片段DNA的量都要比黏末端连接时的用量多，可以增加到5倍以上。2、载体务必去磷酸化处理(许多公司都有这类酶，比如NEB的CIP(点击进入产品页面)。3、如果是PCR生成插入片段，要用产生平端片段的PCR酶(如PFU)，必须磷酸化5'末端，可以磷酸化引物，也可以磷酸化产物，我一般磷酸化产物，这一步最关键也最容易遗漏(请参考T4PNK(点击进入产品介绍)。普通Taq酶PCR以后可以补平再做连接，但是仍然需要磷酸化，当然如果是PCR以后再酶切
