Monoclonal antibody against myelin 2,3-cyclic nucleotide 3-phosphodiesterase (myelin CNPase) Intended Use **Research Use Only (RUO)**
This product is sold for laboratory research use only, not for human or in-vivo use.Clone SMI-91 Form Ascites Fluid Host Mouse Species Reactivity Myelin CNPase (SMI 91) Monoclonal AntibodyHuman, rat, mouse, sheep, dog, pig, bovine IsoType IgG1 Specificity SMI 91 reacts with the 46 kDa and 48 kDa subunits of the 94 kDa myelin CNPase dimer. It detects developing and adult myelin, developing oligodendrocytes and Schwann cells and distinguishes oligodendrocytes from astrocytes, microglia, neurons and other cells in brain sections. The antibody reacts with tissues of human, rat, mouse, sheep, dog, pig or bovine origin. SMI 91 does not react with guinea pig or avian tissues. The combination of SMI 91 with SMI 94 and/or 99 is useful for immunocytochemical studies on the progression of normal and pathologic myelination. For immunocytochemical localization with SMI 91 use of mouse ClonoPAP (Cat. No. SMI-4050L) and high titer goat anti-mouse IgG (Cat. No. SMI-5010C) are recommended. Uses This antibody is effective in immunoblotting (WB), immunocytochemistry, immunohistochemistry (IHC) and ELISA. Suggested Working Dilution The optimal working dilution should be determined for each specific assay condition. - WB: 1:200

Myelin CNPase (SMI 91) Monoclonal AntibodyThe extent of permissible dilution of SMI 91 beyond those recommended for general application depends upon nature and concentration of the antigen examined, species of the antigen, method of fixation and kind of section examined.
Tissue Preparation: SMI 91 reacts on Western blots, paraffin, vibratome or frozen tissue sections and cell cultures. Tissues can be fixed in a variety of paraformaldehyde or formaldehyde-containing fixatives. Post-fixation in cold methanol, methanol/hydrogen peroxide or detergent is necessary for access of SMI 91 to oligodendrocytes and myelin in frozen and thick vibratome tissue sections or in tissue culture. Reaction on formalin-fixed, paraffin-embedded sections may be enhanced by autoclaving deparaffinized sections in distilled water for 10 min (Shin et al, Lab Invest 64:693,1991) or by boiling sections or tissue blocks immersed in tris buffered saline, pH 9.0, in a microwave oven for 15 min (Evers, Uylings, J Neurosci Methods 72:197, 1997).Storage Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody. For long-term storage, keep the antibody at -80°C.
