Neurofilaments, Hypophosphorylated (SMI 35) Monoclonal Antibody IgG1 Specificity 
While other antiphosphoneurofilaments react with neurofilaments of high degrees of phosphorylation, SMI 35 reacts with neurofilaments of low degrees of phosphorylation as well. On two-dimensional immumoblots, it detects a band extending from the phosphorylated neurofilament position at 200kDa (pI 5.1) toward the non-phosphorylated position at 170kDa (pI 6.2). It reacts primarily with neurofilament H and, to a lesser extent, with neurofilament M in most mammalian species, as well as in chicken and frog (Xenopus). This antibody stains thick and thin axons and faintly stains cell bodies. It reveals an especially rich network of thin axons. Other cells and tissues are unreactive except for peripheral axons. In line with the low degree of neurofilament phosphorylation sufficient for reacion with SMI 35, more extensive phosphatase treatment of neurofilaments is necessary for abolition of reaction with this antibody than with any other antibody to phosphorylated neurofilaments. In pathological conditions, SMI 35 frequently detects early changes in neuronal cell body staining. A Golgi-like image of pathologic phosphorylation in neuronal perikarya and their dendritic arborizations has been obtained by ClonoPAP immunocytochemistry with SMI 35 providing a pre-chromatolytic, retrograde demonstration of neuronal connectivities. SMI 35 reacts with occasional intraneuronal tangles in Alzheimers disease. This reaction increases in scope and degree with mild dephosphorylation but is abolished by extensive dephosphorylation. This antibody is effective in immunoblotting, immunocytochemistry and ELISA. This antibody can be used for neurofilament detection in trypsin-treated tissue. Suggested Working Dilution

The extent of permissible dilution of SMI 35 beyond those recommended for general application depends upon nature and concentration of the antigen examined, species of the antigen, method of fixation and kind of section examined.
Tissue Preparation: SMI 35 reacts on Western blots, paraffin, vibratome and frozen tissue sections as well as with neuronal cell cultures. Tissues and cultures can be fixed in a variety of paraformaldehyde or formaldehyde-containing fixatives including Bouins. "Recovery" of antigen in formalin-fixed tissue can be accomplished by autoclaving of deparaffinized sections in distilled water (Shin et al, Lab Invest, 64:693, 1991) or by boiling sections or tissue blocks immersed in tris buffered saline, pH 9.0, in a microwave oven for 15 min (Evers and Uylings, J Neurosci Methods 72:197, 1997). Post-fixation in cold methanol greatly facilitates access of SMI 35 in cryostat or thick tissue sections and in tissue cultures.Notes

Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody. For long-term storage, keep the antibody at -80°C. References
