MTT细胞增殖及细胞毒性检测试剂盒(MTT Cell Proliferation and Cytotoxicity AssayKit)被广泛应用于细胞增殖和细胞毒性的检测。MTT是噻唑蓝(3-(4 , 5-dimethylthiazol-2-yl)-2 , 5-diphenyl tetrazolium bromide),可以被线粒体内的一些脱氢酶还原生成结晶状的深紫色产物甲臜(formazan)。formazan可以被Formazan溶解液完全溶解,然后通过酶标仪测定490nm波长附近的吸光度。细胞增殖速度越快,则吸光度越高;细胞毒性越大,则细胞增殖减慢从而导致吸光度降低。

本试剂盒在经典操作方法的基础上 , 采用了独特的Formazan溶解液, 可以直接溶解formazan,而无需离心去除原有的培养液,故可避免因formazan部分被吸除而引起的操作误差,具有本底低,灵敏度高,线性范围宽,操作简便等特点。
保存条件:
4℃或-20℃保存,需避光保存,一年有效;配置成溶液后需-20℃避光保存。
注意事项:
1. MTT染色液为黄色,需避光保存,长时间光照会导致失效。当颜色变为灰绿色时,请勿使用。MTT染色液在4℃或较低温度情况下会凝固,可以20-25℃水浴温育片刻至全部融解后使用。
2. 由于使用96孔板进行检测,如果细胞培养时间较长,一定要注意蒸发的问题。一方面,由于96孔板周围一圈最容易蒸发,可以采取弃用周围一圈的办法,改加PBS, 水或培养液;另一方面,可以把96孔板置于靠近培养箱内水源的地方,以缓解蒸发。
3. 设空白组(不加细胞,只加培养基),最后比色时,以空白组孔调零。
4.悬浮细胞用此法时,必须经离心后才能吸出上清再加Formazan溶解液。

操作流程:
1. 通常细胞增殖实验96孔板每孔加入100μl 2000个细胞,细胞毒性实验每孔加入100μl 5000个细胞,37℃细胞培养箱培养24h。
2. 加入适当浓度的受试化合物,按照实验要求在细胞培养箱内培养。
3. 之后每孔加入20μl MTT染色液,在细胞培养箱内继续孵育4h。
4. 吸出上清,每孔加入150μl Formazan溶解液,直至 formazan全部溶解。
5. 在490nm测定吸光度。
