This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human HIF-1 alpha / HIF1A (rh HIF-1 alpha / HIF1A; Catalog#11977-H07E; Q16665-1; Arg575-Asn826). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.
HIF-1 alpha / HIF1A抗体 Background
Hypoxia-inducible factor 1-alpha, also known as HIF-1-alpha (HIF1A), ARNT-interacting protein, Member of PAS protein 1, Basic-helix-loop-helix-PAS protein MOP1, PAS domain-containing protein 8, Class E basic helix-loop-helix protein 78 and HIF1A, is a cytoplasm and nucleus protein. HIF-1 alpha/HIF1A contains one basic helix-loop-helix (bHLH) domain, one PAC (PAS-associated C-terminal) domain and two PAS (PER-ARNT-SIM) domains. It is expressed in most tissues with highest levels in kidney and heart. HIF-1 alpha/HIF1A is overexpressed in the majority of common human cancers and their metastases, due to the presence of intratumoral hypoxia and as a result of mutations in genes encoding oncoproteins and tumor suppressors. HIF-1 alpha/HIF1A functions as a master transcriptional regulator of the adaptive response to hypoxia. HIF-1 alpha/HIF1A activates the transcription of over 40 genes, including, erythropoietin, glucose transporters, glycolytic enzymes, vascular endothelial growth factor, and other genes whose protein products increase oxygen delivery or facilitate metabolic adaptation to hypoxia. HIF-1 alpha/HIF1A plays an essential role in embryonic vascularization, tumor angiogenesis and pathophysiology of ischemic disease. HIF-1 alpha/HIF1A interacts with the HIF1A beta / ARNT subunit. Heterodimerization of HIF1A is required for DNA binding. HIF-1 alpha/HIF1A interacts with COPS5. The interaction increases the transcriptional activity of HIF1A through increased stability.
