Produced in BALB/c mice using highly pure recombinant human TNF-α as the immunizing antigen. This antibody was purified from cell culture by Protein G affinity chromatography.

Sandwich
In a sandwich ELISA (assuming 100µl/well), a concentration of 2.0-4.0 µg/ml of this antibody will detect at least 1000 pg/ml of recombinant Human TNF-α when used with PeproTech's biotinylated antigen affinity purified anti-Human TNF-α (500-P31ABt) as the detection antibody at a concentration of approximately 0.5-1.0 µg/ml.
Western Blot
To detect Human TNF-α by Western Blot analysis this antibody can be used at a concentration of 0.5-1.0 µg/ml. When used in conjunction with compatible secondary reagents the detection limit for recombinant Human TNF-α is 0.5-1.0 ng/lane, under reducing conditions and 0.25-0.50 ng/lane, under non-reducing conditions.

To yield one-half maximal inhibition [ND50] of the biological activity of Human TNF-α (0.5 ng/ml), a concentration of < 0.1 μg/ml of this antibody is required.
Immunohistochemistry
This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentrations are 15.0-20.0 μg/ml overnight at 4˚C. An HRP-labeled polymer detection system was used with DAB Chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended. Optimal concentrations and conditions may vary. Description:
TNF-α is a pleiotropic pro-inflammatory cytokine secreted by various cells, including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF family of ligands, and signals through two receptors, TNFR1 and TNFR2. TNF-α is cytotoxic to a wide variety of tumor cells, and is an essential factor in mediating the immune response against bacterial infections. TNF-α also plays a role in the induction of septic shock, autoimmune diseases, rheumatoid arthritis, inflammation, and diabetes. Human and murine TNF-α demonstrate significant cross-species reactivity. TNF-α exists in two forms; a type II transmembrane protein, and a mature soluble protein. The TNF-α transmembrane protein is proteolitically cleaved to yield a soluble, biologically active, 17 kDa TNF-α, which forms a non-covalently linked homotrimer in solution. Recombinant Human TNF-α is a soluble 157 amino acid protein (17.4 kDa) which corresponds to C-terminal extracellular domain of the full length transmembrane protein.
