【背景资料】：In yeast the cytoplasmic cysteine protease Atg4 is responsible for activation of Atg8 by removal of its C-terminal extension to expose the glycine residue essential for its subsequent phospholipid (PE) conjugation. Atg4 is also required for the deconjugation of Atg8 from PE at outer surface of autophagosomes and their precursor membranes, a process required for clustering and fusion with lysosomes, resulting in the recycling of Atg8 and destruction of the autophagosome’s contents. In humans there are four independent Atg4 orthologs, known as autophagins, Atg4A / autophagin-2, Atg4B / autophagin-1, Atg4C / autophagin-3 and Atg4D / autophagin-4, together with at least six Atg8 orthologs (LC3A-C, GABARAP (-L1, L-2)). Of the four Atg4 enzymes only Atg4B has been shown to effectively cleave LC3 and GABARAP precursor proteins as well as to deconjugate them from PE.
