- Murine RAW 264.7 macrophage cells were grown in media containing 100 units/ml penicillin/streptomycin (Invitrogen). Human embryonic kidney 293 cells were grown to 80% confluency and routinely passaged in Dulbecco's modified Eagle's medium containing 10% fetal bovine serum and 100 units/ml penicillin/streptomycin (Invitrogen). Five µg of total RNA from the RAW 264.7 cells were used as templates for the RT-PCR using random hexamers and Superscript Reverse Transcriptase II.

- PCR was used to amplify the 324 bp fibronectin gene. The PCR products were cloned using the TA PCR-cloning system. Cloning was performed in both orientations and accordingly sense and antisense mRNA probes were produced using the T7 promoter of the PCR 2.1 vector. Total RNA was extracted from sciatic nerves and Schwann cell cultures using Trizol. cDNA was then generated using Superscript II reverse transcriptase.
