QS11 is a potent and cell-permeable purine derivative that synergizes with Wnt-3a ligand in the activation of Wnt/β-catenin signal transduction. This compound functions to bind and inhibit the GTPase activating protein of ADP-ribosylation factor 1 (ARFGAP1), and thereby leading to an increase in activated ARF and subsequent β-catenin translocation. QS11 displays potent activity (EC50= 0.5 μM) with little cytotoxicity toward HEK293 and human primary fibroblast cells (≥10 μM, which is QS11's maximum solubility in culture media).QS11 also effectively reduces the migration of metastatic human breast cancer cells [1].
