Introduction Heat shock proteins are molecular chaperones that have an ability to protect proteins from damage induced by environmental factors such as free radicals, heat, ischaemia, and toxins, allowing denatured proteins to adopt their native configuration. Heat shock protein-27 (HSP27) is a member of the small HSP (sHSP) family of proteins and has a molecular weight of approximately 27 KDa. In addition to its role as a chaperone, it has also been reported to have many additional functions. These include effects on the apoptotic pathway, cell movement, and embryogenesis (1). It is suggested that HSP27 may play a key role in resistance to doxorubicin-induced cardiac dysfunction (2). Lower lymphocyte HSP27 levels might be associated with an increased risk of lung cancer (3). HSP27 expression is enhanced in target tissues of diabetic microvascular complications, and changes in circulating serum HSP27 levels (sHSP27) have been reported in patients with macrovascular disease (4).
Principle of the Assay The AssayMax Human HSP27 ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for detection of human HSP27 in plasma, serum, milk, tissue extract, and cell culture samples. This assay employs a quantitative sandwich enzyme immunoassay technique that measures human HSP27 in less than 5 hours. A polyclonal antibody specific for human HSP27 has been pre-coated onto a 96-well microplate with removable strips. HSP27 in standards and samples is sandwiched by the immobilized antibody and biotinylated polyclonal antibody specific for HSP27, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.
