| 描述 | Dapolsertib (SEL24-B489) is an orally bioavailable and potent PIM and FLT3-ITD inhibitor, inhibiting PIM1, PIM2, and PIM3, decreasing PIM substrate phosphorylation, and inducing dose-dependent apoptosis in leukemia cells. |
| 体外活性 | 在 MOLM-13 细胞和 MV4-11 细胞中，观察到 Dapolsertib 处理后细胞周期的剂量依赖性中断，特别是 S 期的明显缺失，伴随着 PARP 的切割和凋亡。 Dapolsertib 能对 S6 （s235 /236）产生深远的抑制作用，但对 PI3K/mTOR 信号通路的影响很小。 Dapolsertib 抑制 STAT5 （Ser 726）并降低 MCL1 的表达，而没有抑制 c-MYC 丰度或诱导 PARP 切割。 细胞活力测定： AmL 细胞系 AZD1208， AC220 和 AraC 浓度：0 ~ 10 μM 孵育时间：72小时 结果：活力下降。[1] |
| 体内活性 | Dapolsertib （25-100 mg/kg，口服）在AmL体内模型中显示出活性。[1] Dapolsertib 在低/亚微摩尔浓度下诱导 DLBCL 细胞系凋亡，并在异种移植模型中表现出活性。[2] 动物模型：携带 MV-4-11 肿瘤（FLT3-ITD+）的 SCID/beige 小鼠。剂量：50mg/kg、75 mg/kg、100mg/kg。用法：口服，每日两次。结果：显著的剂量依赖性肿瘤减少（50、75 和 100 mg/kg 每日剂量分别为 67%、74% 和 82% 的肿瘤生长抑制（TGI））。[1] |
