| 描述 | Dabuzalgron (Ro 115-1240) is an orally active selective alpha-1A adrenergic receptor agonist used to treat urinary incontinence. Dabuzaron prevents cardiotoxicity caused by doxorubicin by maintaining mitochondrial function. |
| 体外活性 | Dabuzaron处理以剂量依赖性方式诱导 ERK 磷酸化，其EC50为4.8 μM。ERK1/2的激活有助于Dabuzaron的心脏保护作用。Dabuzaron（10 μM；4小时）保护NRVM免受Doxorubicin （DOX）诱导的细胞死亡。Dabuzaren（10 μM；4小时）激活α1A-AR减轻了DOX对线粒体膜电位的有害影响，并消除了对线粒体损伤的凋亡反应中关键元素的激活。 |
| 体内活性 | Dabuzaron（10μg/kg；口服管饲；每日两次；连续7天；C57B16J野生型或α1A-AR基因敲除小鼠）通过激活α1A-AR，可预防Doxorubicin 引起的心脏毒性。Dabuzaron可维持DOX处理小鼠心脏中的ATP含量，减少氧化应激，防止有关线粒体功能转录的降低，并上调PGC1α表达。 |
