| 描述 | Oclacitinib maleate (PF-03394197 maleate) is a novel inhibitor of JAK family members with IC50 ranging from 10 to 99 nM and JAK1-dependent cytokines with IC50 ranging from 36 to 249 nM, which did not inhibit a panel of 38 non-JAK kinases. |
| 体外活性 | Oclacitinib在10至99nM的浓度范围内抑制JAK家族成员50%，且不抑制38种非JAK激酶。Oclacitinib对JAK1的抑制作用最强。Oclacitinib还能够抑制依赖JAK1的细胞因子的功能，这些细胞因子涉及过敏、炎症以及瘙痒。Oclacitinib对不激活JAK1酶的细胞因子具有最小的影响[1]。 |
| 体内活性 | Oclacitinib通过口服方式给予，剂量为0.4-0.6mg/kg，每天两次，安全有效地控制了与过敏性皮炎相关的瘙痒症状。Oclacitinib在24小时内开始缓解瘙痒，并在整个治疗期间持续有效，超过70%的受治疗犬在第7天实现了瘙痒程度>50%的减少[2]。 |
