| 描述 | Tamoxifen is an orally active selective estrogen receptor modulator (SERM) that acts as an estrogen antagonist in breast cells and an agonist in bone, liver, and uterine cells. It can be used to induce gene knockout and liver injury models in mice, and also exhibits multiple biological activities, including activation of Hsp90, induction of autophagy and apoptosis, and inhibition of EBOV and MARV viral infections. |
| 体外活性 | 方法：人乳腺癌细胞 MCF-7 用 Tamoxifen (0.25-4 μM) 处理 24 h，使用 CCK-8 方法检测的细胞活力。 结果：Tamoxifen 以剂量依赖性的方式显著抑制 MCF-7 细胞的增殖。[1] 方法：ER 阴性乳腺癌细胞 SK-BR3、MDA-MB-453、MDA-MB-468、MDA-MB-231 和 HCC-1937 用 Tamoxifen (1-10 μM) 处理 24-36 h，使用 Flow Cytometry 方法检测细胞凋亡情况。 结果：Tamoxifen 以剂量和时间依赖的方式诱导 MDA-MB-231、MDA-MB-468、MDA-MB-453 和 SK-BR3细胞凋亡，而在 HCC-1937 细胞中未观察到明显的凋亡作用。[2] |
| 体内活性 | 方法：为检测体内抗肿瘤活性，将 Tamoxifen (100 mg/kg) 口服给药给携带 ER 阴性乳腺癌肿瘤 MDA-MB-468 或 HCC-1937 的 NCr athymic nude 小鼠，每周三次，持续四-五周。 结果：Tamoxifen 显著抑制 MDA-MB-468 肿瘤生长，而 HCC-1937 肿瘤生长不受影响。[2] 方法：为测试对小鼠行为的影响，将 Tamoxifen (75 mg/kg in 10% ethanol+90% sunflower seed oil) 腹腔注射给 C57BL/6 小鼠，每天一次，持续七天。 结果：Tamoxifen 会影响小鼠的运动活动、社交和焦虑。[3] |
