| 描述 | Biricodar dicitrate (VX-710) is a potent MDR inhibitor with no anticancer activity and can be used to study prostate cancer. |
| 体外活性 | Biricodar 对 P-glycoprotein (Pgp) 和 MRP-1 均显示出活性，通过增强药物吸收和保留同时逆转由野生型 BCRP (BCRPR482) 调控的药物抗性。在 Pgp 表达的 8226/Dox6 细胞中，Biricodar 分别将 mitoxantrone 和 daunorubicin 的吸收增加了 55% 和 100%，其保留量分别增加了 100% 和 60%，并将它们的细胞毒性分别提高了 3.1 倍和 6.9 倍。此外，Biricodar 对 HL60/Adr（MRP-1）和8226/MR20（BCRP(R482)）细胞显示出类似效果，但对 MCF7 AdVP3000 细胞（BCRP(R482T)）影响不大。VX-710，一种非大环内酯类 pipecolinate 衍生物，通过与 FK506 受体蛋白结合，已被证明可在包括骨髓瘤、黑色素瘤、癌症和白血病在内的多种多药耐药细胞中恢复敏感性。同时，Biricodar 有效抑制了 P-glycoprotein 通过 [3H]azidopine 或 [125I]iodoaryl azido-prazosin 的光亲和标记，EC50 值分别为 0.75 和 0.55 μM。 |
