| 描述 | Robinin (NSC-9222) belongs to natural products, serving as a TLR2 inhibitor with cell permeability. Robinin exerts anti-inflammatory and antitumor activities through inhibiting the TLR2-PI3K-AKT pathway, and can be used for experimental research related to pancreatic cancer. |
| 体外活性 | 方法;采用小鼠原代软骨细胞，经IL-1β（10 ng/mL）处理12 h构建损伤模型后，以10 μg/mL Robinin孵育24 h。 结果：CCK-8及流式细胞术显示，Robinin可逆转IL-1β所致的细胞活力下降，并抑制细胞凋亡；Western blot进一步证实其上调Bcl-2、下调Bax与C-caspase3表达，发挥保护作用。[1] 方法：采用H9c2大鼠心肌细胞，以50 μM Robinin预处理24 h，再分别加入40 μM H₂O₂或5 μM多柔比星诱导损伤。 结果：MTT、AO/EB及Hoechst染色显示，Robinin可提高细胞活力，减少凋亡；DCFH-DA及流式显示其降低活性氧水平，并增强抗氧化酶活性，抑制内质网应激相关基因与蛋白表达。[2] |
| 体内活性 | 方法：在高胆固醇血症大鼠模型中，通过2%胆固醇饲料诱导8周后，于第6周起腹腔注射Robinin（50 mg/kg，每两天一次，溶于DMSO，对照为溶剂），连续给药2周，随后行离体心脏缺血再灌注损伤。 结果：Robinin可改善心功能，抑制线粒体通透性转换孔开放，减轻心肌组织病理损伤，并上调心肌Akt、Nrf2、HO-1及NQO1表达，下调GSK3β与Fyn水平。[3] |
