| 描述 | Fisetin belongs to a group of natural flavonols with a variety of biological activities, including anti-inflammatory, antioxidant, anti-tumor, anti-aging and neuroprotective effects. |
| 体外活性 | 方法: 人胰腺癌细胞 PANC-1 用 Fisetin (25-400 µM) 处理 24-48 h，通过 CCK-8 assay 检测细胞活力。 结果: 低浓度的 Fisetin (25-50 µM) 并没有显著抑制 PANC-1 细胞的存活率，高浓度抑制。[1] 方法: HeLa 细胞用 Fisetin (20-50 µM) 处理 24-48 h，通过 Flow cytometry 检测细胞周期。 结果: 用 20/30/50 µM Fisetin 处理的细胞导致 G2/M 期细胞大量积聚。在 24 h 内，G2/M 停滞细胞的比例分别从 10.1% 增加到 16.2%、18.9% 和 25.1%，而在 48 h 内，G2/M 阻滞细胞比例分别增加到 30.9%、36.2% 和 56.2%。同时，在 48 h 时，50 µM 的 Fisetin 使很大一部分细胞处于 Go/G1 期。[2] |
| 体内活性 | 方法: 为检测体内抗肿瘤活性，将 Fisetin (300 mg/kg) 腹腔注射给携带 PANC-1 异种移植物的 BALB/c nude 小鼠，每两天一次，持续 20 天。 结果: Fisetin 治疗的小鼠肿瘤大小显著减小。Fisetin 治疗组的增殖相关蛋白增殖细胞核抗原 (PCNA) 显著降低。Fisetin 治疗降低了细胞生长和增殖相关蛋白的表达，包括 PCNA、Ki67 和 磷酸化组蛋白 H3。[1] |
