【原 理】 本实验采用磺酸型阳子交换树脂（732型）分离酸性氨基酸（天冬氨酸Asp pI=2.97）和硷性氨基酸(赖氨酸Lys pI=9.74)的混合液。在pH5.3条件下，因为低于Lys的pI值，Lys可解离成阳离子挂在树脂上；高于Asp的pI值，则Asp可解离为阴离子，不能被树脂吸附而直接流出色谱柱，在pH 12条件下，因高于Lys的pI值，Lys又解离为阴离子从树脂上被交换下来，这样通过改变洗脱液的pH值可使它们被分别洗脱而达到分离
