| 描述 | Zardaverine (BY 290) is an orally available, selective and potent PDE3/4 inhibitor with bronchodilator activity and anti-hepatocarcinogenic activity.Zardaverine's action in platelets is mediated by PDE III isoenzymes and can be used in studies of acute renal failure and chronic airflow obstruction. |
| 体外活性 | Zardaverine在0至30µM浓度范围内，连续作用72小时对人类肝细胞癌(HCC)细胞的生长具有选择性抑制作用[3]。其抗肿瘤活性与细胞周期相关蛋白的调控密切相关，但与PDE3/4抑制作用无关[3]。在0.1µM浓度作用24小时时，Zardaverine能够选择性地引导HCC细胞进入G0/G1期阻滞，并且使细胞周期相关蛋白失调[3]。此外，Zardaverine在Bel-7402和SMMC-7721细胞中以时间和浓度依赖的方式诱导凋亡，使用的浓度包括0.01、0.03、0.1、0.3和1µM（持续48小时），以及0.3和1µM（作用24、36、48、60和72小时）[3]。 |
