| 描述 | Cerivastatin is an orally active and highly effective HMG-CoA reductase inhibitor with anticancer and lipid-lowering effects. Cerivastatin can reduce low-density lipoprotein cholesterol levels, inhibit the proliferation and invasion of MDA-MB-231 cells, and can be used to study primary hyperlipidemia. |
| 体外活性 | Cerivastatin可诱导 MDA-MB-231 细胞增殖的剂量依赖性下降（25 毫微克/毫升时抑制率可达 40%）。相比之下，Cerivastatin并没有明显改变 MCF-7 细胞的增殖。流式细胞术分析表明，Cerivastatin可诱导细胞周期在处理 36 小时后停滞在 G 1 /S 期（在Cerivastatin处理的细胞中占 67.1%）。[1] |
| 体内活性 | 曲美替尼和 Cerivastatin 最初溶于二甲基亚砜，然后在生理盐水中稀释成最终浓度为 0.5% 的低聚异麦芽糖和 0.05% 吐温-80 的溶液。在高度免疫缺陷的 NOD.Cg-Prkdcscid Il2rgtm1wjl/SzJ 小鼠体内接种原代单体BAP1突变人类UM细胞系 UPMM3 的异种移植，以验证曲美替尼和 Cerivastatin 联合疗法对体内UM细胞的影响。皮下注射 UPMM3 细胞一周后，当肿瘤可触及时，每组四只小鼠分别接受溶剂、曲美替尼（1毫克/千克，每只，三天/周）、Cerivastatin （2毫克/千克，每只，三天/周）或曲美替尼和 Cerivastatin 治疗57天（至第64天）。治疗结束后进行为期 15 天的观察。与单用曲美替尼治疗的小鼠相比，添加 Cerivastatin 可显著增强对肿瘤生长的抑制作用（p = 0.03）。[4] |
