Prucalopride is a high affinity agonist for 5-HT4 receptor selection. Its pK(i) values for the human 5-HT(4a) and 5-HT(4b) receptor, are 8.60 and 8.10, respectively. Furthermore, it caused relaxation of a rat oesophagus preparation (pEC(50)=7.81+/-0.17) in a 5-HT(4) receptor antagonist sensitive manner3. The viability of SVG p12 cells was inhibited by Prucalopride beyond 50 μM. Prucalopride(10 μM ) inhibited glioma cells growth, its IC50 value was 9.942 ± 0.346 μM4. Prucalopride maximally increased ACh(acetylcholine) and histamine levels in the prefrontal cortex at 5 and 10 mg/kg5.
