| 检测原理 | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human NUCB1. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human NUCB1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human NUCB1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human NUCB1 in the samples is then determined by comparing the OD of the samples to the standard curve. |

相表面上进行吸附-解吸附-再吸附，并在此过程中逐渐被复性，形成与天然蛋白质构象相同的蛋白质，并流出色谱柱。HIC固定相是从高盐溶液中吸附变性蛋白质，且与变性剂瞬时分离，不仅大大降低了蛋白质间的聚集作用，还因固定相在分子水平上为变性蛋白提供里很高的能量，使水化的变性蛋白质瞬时失水，并形成局部结构以利于蛋白质从疏水核开始折叠。HIC在蛋白质复性的同时还能与其它杂蛋白进行很好的分离，且HIC柱便宜、快速，故有很好的发展潜力。 5、 反胶束复性法[27]：由于蛋白质在反胶束内水相中可以保持其构象和活性
