检测原理

微孔板预包被抗大鼠去甲肾`上`腺`素的捕获抗体。加入样本或稀释的标准品后，NE分子与捕获抗体形成复合物。随后加入生物素化检测抗体和链霉亲和素‑HRP，构建抗体‑抗原‑抗体‑酶复合物。经洗涤后加入TMB底物，在HRP催化下显色（蓝），加终止液转为黄色，用酶标仪在 450 nm 读取光密度（OD），OD 与样本中NE浓度呈正比。

Product Overview

This kit is a core product of the EliKine™ series, specifically designed for the quantitative detection of Noradrenaline (NE/NA/NAd/Norepi) in rat serum, plasma, tissue, cell supernatant, and other liquid samples. It employs a double-antibody sandwich method to achieve highly sensitive and specific ELISA detection.

Detection Principle

The microplate is pre-coated with a capture antibody against rat noradrenaline. After adding samples or diluted standards, NE molecules form complexes with the capture antibody. Subsequently, a biotinylated detection antibody and Streptavidin-HRP are added to form an antibody-antigen-antibody-enzyme complex. After washing, TMB substrate is added, which develops a blue color catalyzed by HRP. The reaction is stopped by adding stop solution, turning the color yellow. The optical density (OD) is measured at 450 nm using a microplate reader, and the OD value is proportional to the NE concentration in the sample.

Suitable Samples

Serum, plasma, tissue homogenate, cell culture supernatant, and other related liquid samples. Duplicate or triplicate wells are recommended to reduce experimental error.

Precautions for Use

• Do not mix reagents from different lots or expired reagents.
• Allow all reagents to reach room temperature (equilibrate for 30 minutes) before use.
• Use disposable pipette tips to prevent cross-contamination during operation.
• Return unused strips to the original packaging immediately, seal, and store at 4 °C.
• Thorough mixing significantly affects results; low-speed shaking or manual shaking every 10 minutes is recommended.
• Duplicate or triplicate wells are recommended for samples and standards.
