检测原理

采用竞争法ELISA：

1. 微孔板已预包被抗小鼠皮质醇抗体。
2. 加入样本或标准品后，皮质醇分子与抗体竞争结合位点。
3. 随后加入HRP标记的皮质醇，未被样本占据的位点被标记分子占据。
4. 洗涤除去未结合组分。
5. 加入TMB底物，显色强度与孔内游离HRP标记分子量成正比，因而与样本中皮质醇浓度呈负相关。
6. 使用Stop Solution终止反应，使用酶标仪在450 nm测光密度（OD），依据标准曲线计算样本浓度。

实验操作要点

Assay Principle

Utilizes a competitive ELISA method:

1. The microplate is pre-coated with an anti-mouse cortisol antibody.
2. After adding the sample or standard, cortisol molecules compete for antibody binding sites.
3. HRP-conjugated cortisol is then added, occupying sites not bound by sample cortisol.
4. Unbound components are removed by washing.
5. TMB substrate is added; the color intensity is proportional to the amount of free HRP-conjugate, and thus inversely proportional to the cortisol concentration in the sample.
6. The reaction is stopped using Stop Solution, and the optical density (OD) is measured at 450 nm using a microplate reader. Sample concentration is calculated based on the standard curve.
