TrailBlazerTM StarBrightTM Dye Label is the second part of a two-kit antibody labeling system. Together with the TrailBlazer Tag kit it will allow you to conjugate an antibody to superior StarBright Dyes using the innovative SpyTag and SpyCatcher technology. They provide the benefits of using StarBright Dyes in flow cytometry and western blots for your antibody of choice.
First, the SpyTag is attached to an antibody using the TrailBlazer Tag Kit. This antibody, now with a SpyTag, is then mixed with a StarBright Dye labeled SpyCatcher using the Label Kit. A spontaneous reaction occurs between the SpyTag and SpyCatcher resulting in an irreversible covalent isopeptide bond. The result is a highly stable StarBright Dye labeled antibody, which should be stored at +4°C in the dark, and when stored correctly will be stable for up to 12 months.

In flow cytometry we suggest using 5µl of the undiluted labeled antibody to stain 106 cells in 100µl. However, we highly recommend that you titrate the antibody to determine the optimal amount of antibody for your specific experiment.

Flow Cytometry
Use 5ul of the suggested working dilution to label 106 cells in 100ul. Best practices suggest a 5 min centrifugation at 6,000g prior to sample application.
Instructions For Use

- Add the SpyTagged antibody product from TrailBlazer Tag Kit to the StarBright Dye-Catcher vial. Mix and incubate for 30 min at room temperature or overnight at +4°C.
- Add the sample to the Spin Column. Vortex briefly and mix on an end-over-end rotary mixer at room temperature for 60 min.
- Break open the bottom of the Spin Column and insert the column into the 2 ml amber collection tube. Slightly loosen the cap of the Spin Column and centrifuge at 1500 x g for 2 min. Retain the eluate, containing the label antibody.
- Prepare Quenching Reagent by adding 5 µl of ultrapure water.
- Add 4 µl of prepared Quenching Reagent and 45 µl of Storage Buffer to the labeled antibody. Vortex briefly to mix.
Antibodies are now labeled and ready to use. Store at +4°C in the dark for up to 1 year.
