Identification and enumeration of CD201 (EPCR)+ cells by flow cytometryClone 1560 recognizes the mouse CD201 antigen, a single-pass type I membrane protein which is also known as endothelial protein C receptor (EPCR). CD201 is strongly expressed in the endothelial cells of arteries and veins in heart and lung, and less intensely in capillaries in the lung and skin. It is also expressed on a subset of neutrophils, monocytes, and several tumor cells. It was demonstrated that murine CD201 is expressed at high levels within the bone marrow by hematopoietic stem cells (HSC). Bone marrow cells isolated on the basis of CD201 expression alone are highly enriched for hematopoietic reconstitution activity, showing levels of engraftment in vivo. The major role of CD201 is anticoagulation. CD201 binds to protein C with high affinity and augments the activation of protein C by the thrombin-thrombomodulin complex. Another important functional aspect of CD201 is its role in many pathophysiologic processes, such as inflammation responses to infection, trauma, hematopoiesis, and autoimmune response. Deletion of the CD201 gene in mice has been reported to lead to embryonic lethality before embryonic day 10. Overexpression of CD201 results in resistance to formation of thrombin in response to a procoagulant stimulus and decreased susceptibility to endotoxin-induced septic shock. Mice with severe CD201 deficiency have been reported to have no measurable influence on an arterial thrombosis model but have a more severe coagulation response to endotoxemia.
