检测原理

CheKine™ Exo-β-1,4-glucanase (C1) Activity Assay Kit (Micro 1,4-β-D-Glucan Cellobiohydrolase Activity Assay Kit) is a micro-assay kit based on the 3,5-dinitrosalicylic acid (DNS) method, designed for the quantitative analysis of C1 enzyme activity in various biological samples. This kit accurately measures C1 enzyme activity by detecting the amount of reducing sugars generated from the enzymatic degradation of microcrystalline cellulose catalyzed by C1.
Assay Principle
Exo-β-1,4-glucanase (C1, EC 3.2.1.91) catalyzes the hydrolysis of the non-reducing terminal β-1,4-glycosidic bonds in cellulose chains, releasing cellobiose and glucose. This kit utilizes a colorimetric reaction where the reducing sugars produced by the C1 enzymatic reaction react with 3,5-dinitrosalicylic acid to form a brown-red colored compound, which exhibits a characteristic absorption peak at 540 nm. Within a certain concentration range, the rate of change in absorbance at 540 nm is directly proportional to C1 enzyme activity.
