      Specificity Signet’s anti-human spectrin (Clone 17C7) is developed against the alpha I spectrin SH3 domain (Xu et al., 2000). Clone 17C7 is a specific marker for human, rat and mouse alpha I spectrin. It reacts with erythroid spectrin in red blood cells and in cerebellum. It does not crossreact with alpha II (nonerythroid) spectrin SH3 domain or recombinant human fodrin SH3 domain. In cerebellar cell cultures, clone 17C7 labels cell bodies consistent with localization of erythroid spectrin. In NIH 3T3 cells, clone 17C7 reacts with a 200 kDa protein which is a candidate marker of endocytic vesicles called macropinosomes (Xu et al., 2000 & 2001, Lamaze & Schmid, 1995).

      This antibody was developed against a recombinant alpha I spectrin (erythroid spectrin) SH3 domain. It was subsequently purified via ammonium sulfate precipitation.
      Uses Alpha-I Spectrin Monoclonal Antibody (SIGNET)This antibody is effective in immunoblotting, immunofluorescence (IF), immunohistochemistry (IHC) and ELISA.
      Suggested Working Dilution The optimal working dilution should be determined for each specific assay condition.
      • Western blot: 1:1,000- 1:10,000
      • IF: 1:100-1:500
      • IHC: 1:10-1:50 (Frozen only)
      • ELISA: 1:100,000-1:1,000,000
      Storage Store at -20°C or below. Upon initial thawing, apportion into working aliquots and store at -20°C or below. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody. Do not store in frost-free freezers.
