This product is sold for laboratory research use only, not for human or in-vivo use.Form Affinity Purified Antibody Host Rabbit Species Reactivity Nicastrin C-Terminal (Nicastrin/C-Term) Polyclonal Antibody, PurifiedHuman, Mouse [Ab] 0.35 mg/mL Specificity The gamma secretase is a multiprotein complex consisting of presenilin, nicastrin, Aph-1 and Pen-2; all four proteins are necessary for full proteolytic activity. The gamma secretase complex catalyzes cleavage of a number of integral membrane proteins including Amyloid precursor protein, Notch, Erb4 and E-cadherin. The results of several recent studies indicate that the functional gamma secretase activity mostly arises from a low abundance very high molecular weight complex (>670 kDa). These data suggest that additional proteins may be present in these functional complexes or the functional complex is a higher order multimer of presenilin, aph-1, pen-2 and nicastrin.
The antibody was raised against the C-terminal fragment of nicastrin corresponding to a peptide containing residues 691-709 [C]-NAKADVLFIAPREPGAVSY.The peptide was coupled to KLH and affinity-purified (ammonium sulfate precipitated IgG applied to columns with peptides coupled to Sulfo-link).
The PRB-364P was raised against the Nicastrin (N-terminal) and has a similar affinity and specificity to the PRB-365P. Both these nicastrin antibodies recognize transfected and endogenous proteins.Uses Nicastrin C-Terminal (Nicastrin/C-Term) Polyclonal Antibody, PurifiedThis antibody is effective in immunoblotting (WB), immunocytochemistry (ICC) and immunoprecipitation (IP). Suggested Working Dilution The optimal working dilution should be determined for each specific assay condition. - WB: 1:1,000

Storage Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody.
