Background Knowledge

CD28 (Cluster of Differentiation 28) is a key molecule expressed on T cells that provides co-stimulatory signals. It consists of an Ig V-like extracellular domain, a transmembrane domain, and an intracellular domain, existing on the cell surface in the form of a disulfide-linked homodimer or monomer. CD28 is the receptor for B7.1 (CD80) and B7.2 (CD86). When activated by Toll-like receptor ligands, the expression of B7.1 in antigen-presenting cells is upregulated, and the expression of B7.2 is constitutive. It is the only B7 receptor constitutively expressed on naive T cells. In addition to TCR stimulation, stimulation through CD28 can provide a powerful co-stimulatory signal for T cells to produce various interleukins, such as IL-2 and IL-6. Its intracellular domain contains key residues for effective signal transduction and also contains a proline-rich motif that binds to proteins with SH3 domains, and can stimulate eosinophils, releasing IL-2, IL-4, IL-13, and IFN-γ upon binding.
