Biochem/physiol Actions
Fibronectin Type III Connecting Segment (IIICS) is responsible for melanoma cell adhesion.[1] It is the predominant binding site of integrin α4β1, which is mediated through the aspartic acid residue present at position 21.[2] Studies in chicken show that alternative splicing in this region during development of the peripheral nervous system, is essential for the control cell-cell adhesion.[3]
General description
Fibronectin is composed of two cell adhesion domains namely, the central domain for cell-binding made of adhesion-recognizing tetrapeptide, and the alternatively spliced type III connecting segment (IIICS).[1] It contains a tetrapeptide region, from amino acid 90-109, called CS5, which is essential for adhesion mediated by IIICS. Both CS5 and CS1 mediate melanoma cell spreading, when stimulated by immunoglobulin (Ig)G, with CS1 exhibiting higher activity.
