Human Anti-Evolocumab Antibody, clone AbD40410, is a paratope specific, inhibitory antibody that specifically recognizes the monoclonal antibody drug evolocumab. It does not recognize the drug target, human proprotein convertase subtilisin/kexin type 9 (PCSK9), nor evolocumab in complex with PCSK9. The antibody can be used to measure free evolocumab in patient samples.
The monovalent Fab format of this antibody, clone AbD40410ad (TZA008) is tagged with a SpyTag2 at the C-terminus of the Fab antibody heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour.

A pair of anti-evolocumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. This antibody is recommended as the detection antibody when in the bivalent HRP conjugated format, clone AbD40410pap (TZA008P) paired with an anti-evolocumab antibody in monovalent Fab format, clone AbD40406ad (TZA007), as the capture antibody.
Evolocumab (Repatha) is a fully human IgG2 lambda antibody developed using transgenic mouse technology. It has been approved for treating selected patients with high cholesterol, and for the prevention of heart attack and stroke. The drug prevents the PCSK9 protein binding to the low density lipoprotein receptor (LDLR), thereby increasing the number of LDLRs available and lowering LDL levels.
