背景介绍
Tom22 is a central receptor component of the translocase of the outer membrane of mitochondria responsible for the recognition and translocation of cytosolically synthesized mitochondrial preproteins. It is showed in UniProt that phosphorylation is performed at three positions of this protein which may increase the molecular weight. Catalog#66562-1-Ig recognises the 22 kDa protein larger than the calculated MW which is 15 kDa. Besides, it has been reported the MV of TOMM22 is 22 kDa (PMID:19285029).

文献和实验的观点： 正常的，自己配的PBS高压后是会有沉淀的。所以你可以用0.22um的滤器过滤的。 我们一般是先配10×的PBS，用0.22um的滤器过滤后储存。待需要时，稀释成1×的，然后高压，感觉好一点。 丁香园网liujian的观点： 除了与试剂有关外，与水也有很大的关系，不知道楼主用的是什么水？有一次我们实验室的纯水仪的虑芯该换了，用这样的水配出来的PBS高压时就很容易出现沉淀！然后我换了用蒸馏器烧的三蒸水后就没有沉淀了！ 后来我们干脆就配成5×或10×的常温保存，用灭菌3蒸水稀释后再用
PBS是最普遍不过的实验室缓冲液，但其配方各异。以下是我的总结： 母液的配制： 0.2M Na2HPO4：称取 71.6g Na2HPO4-12H2O，溶于 1000ml 水 0.2M NaH2PO4：称取 31.2g NaH2PO4-2H2O，溶于1000ml 水 各种浓度PB(pH=7.4)的配制： 先配 0.2M PB (pH=7.4，100ml)：取19ml 0.2mol/L的 NaH2PO4， 81ml 0.2mol
7 ·2H 2 O：分子量294.12，0.1 mol/L溶液为29.41克/毫升。 6. 乙酸–乙酸钠缓冲液（0.2 mol/L） Na 2 Ac·3H 2 O分子量 = 136.09，0.2 mol/L溶液为27.22克/升。 7. 磷酸盐缓冲液 （1）磷酸氢二钠–磷酸二氢钠缓冲液（0.2）
