Phospho-Tuberin (Ser939 plays a pivotal role in regulating the activity of the TSC1/TSC2 complex, a critical inhibitor of mTORC1 signaling pathway, which governs cell growth, proliferation, and metabolism. Phosphorylation at Ser939 is primarily mediated by the serine/threonine kinase Akt in response to growth factors and insulin signaling. This modification facilitates the binding of Tuberin to 14-3-3 proteins, leading to the inhibition of the TSC1/TSC2 complex's GTPase-activating protein (GAP) activity toward the small GTPase Rheb.
