PTEN (Phosphatase and Tensin Homolog) is a critical tumor suppressor protein that plays a key role in regulating cell growth, division, and survival. It acts primarily by dephosphorylating phosphoinositides, particularly PIP3 (phosphatidylinositol (3,4,5)-trisphosphate), which is a second messenger involved in activating signaling pathways that promote cell growth and survival. PTEN's action leads to the reduction of AKT signaling, a pathway crucial for cell cycle progression, metabolism, and anti-apoptotic processes. By regulating the PI3K/AKT pathway, PTEN helps maintain genomic stability, prevents uncontrolled cell proliferation, and suppresses tumorigenesis. Mutations or deletions of PTEN are commonly found in a wide range of cancers, including breast, prostate, endometrial, and brain cancers, making PTEN a significant target for cancer research and therapeutic development.
