Enhancer of Zeste Homolog 2 (EZH2) is a histone methyltransferase enzyme encoded by the EZH2 gene. It serves as the catalytic subunit of the Polycomb Repressive Complex 2 (PRC2), which mediates the tri-methylation of histone H3 at lysine 27 (H3K27me3), leading to chromatin compaction and transcriptional repression of target genes. EZH2 plays a pivotal role in regulating gene expression during embryonic development, stem cell maintenance, and cell differentiation. Its dysregulation, through overexpression or mutation, has been implicated in various cancers, including prostate, breast, and melanoma, by silencing tumor suppressor genes. Beyond its role in cancer, mutations in EZH2 are associated with developmental disorders such as Weaver syndrome, characterized by overgrowth and advanced bone age.
