Activating Transcription Factor 4 (ATF4) is a stress-responsive transcription factor that plays a pivotal role in cellular adaptation to various environmental and physiological challenges. Belonging to the basic leucine zipper (bZIP) family, ATF4 regulates gene expression in response to stressors such as amino acid deprivation, oxidative stress, and endoplasmic reticulum (ER) stress. It is a central component of the integrated stress response (ISR), modulating genes involved in amino acid metabolism, redox homeostasis, and apoptosis. ATF4's activity is tightly regulated at the translational level through upstream open reading frames (uORFs) in its mRNA, allowing for rapid protein synthesis during stress conditions. Beyond stress responses, ATF4 influences osteoblast differentiation, tumor progression, and neurodegenerative processes, underscoring its significance in both health and disease contexts.
