Hormone-sensitive lipase (HSL) is a crucial intracellular enzyme that regulates lipid metabolism by hydrolyzing stored fats into usable energy. Encoded by the LIPE gene, HSL is primarily active in adipose tissue, where it breaks down triglycerides into free fatty acids and glycerol during lipolysis. This process is hormonally regulated: catecholamines and adrenocorticotropic hormone (ACTH) activate HSL, while insulin inhibits it, balancing energy mobilization and storage. HSL is expressed in steroidogenic tissues like the testes, where it hydrolyzes cholesteryl esters to release free cholesterol, a precursor for steroid hormone synthesis. HSL's broad substrate specificity also includes diacylglycerols, monoacylglycerols, and retinyl esters, highlighting its versatile role in lipid homeostasis. Due to its central role in energy balance, HSL is a significant focus in research areas related to metabolic disorders, including obesity, insulin resistance, and type 2 diabetes.
