SLC19A1 is a solute carrier protein and antiporter that plays a crucial role in nutrient transport and cellular homeostasis. It mediates the import of reduced folates and certain cyclic dinucleotides, driven by the export of organic anions. This secondary active transporter has a high affinity for N5-methyltetrahydrofolate, the predominant circulating form of folate, and is essential for folate absorption in tissues like the liver, kidneys, and intestines. In addition to folates, SLC19A1 also imports immunoreactive cyclic dinucleotides, such as cyclic GMP-AMP (2'-3'-cGAMP), an immune messenger produced in response to DNA viruses in the cytosol. This suggests a role in triggering immune responses, particularly in the context of viral infections. SLC19A1 has also been implicated in the import of methotrexate, an antifolate drug commonly used in cancer chemotherapy, indicating its potential impact on drug efficacy. Mechanistically, SLC19A1 functions by exporting intracellular organic anions down their concentration gradients to facilitate the uptake of its substrates, thus contributing to vital cellular processes such as DNA synthesis, repair, and methylation. Dysregulation of SLC19A1 is linked to metabolic disorders, nutrient deficiencies, and may influence chemotherapy outcomes, making it an important target for research in nutritional science, oncology, and immunology.
