Mitochondrial Rho GTPase 1 (RHOT1), also known as MIRO1, is a mitochondrial outer membrane protein that regulates mitochondrial transport, calcium signaling, and quality control. It anchors mitochondria to motor proteins like kinesin and dynein via TRAK adaptors, facilitating their movement along microtubules . RHOT1 contains two GTPase domains and EF-hand motifs that bind calcium, enabling it to respond to intracellular calcium levels and halt mitochondrial movement when necessary . It also plays a role in mitophagy by interacting with PINK1 and Parkin to remove damaged mitochondria . Mutations in RHOT1 have been linked to Parkinson’s disease and cancer progression.
