Histone deacetylase 4 (HDAC4) is a member of the class IIa histone deacetylase family, playing a pivotal role in regulating gene expression through chromatin remodeling. Unlike some other HDACs, HDAC4 does not bind directly to DNA; instead, it is recruited to specific gene promoters via interactions with transcription factors such as MEF2C and MEF2D. Once localized to chromatin, HDAC4 removes acetyl groups from histone tails, leading to chromatin condensation and transcriptional repression. HDAC4 is involved in various biological processes, including skeletal and cardiac muscle development, neuronal survival, and synaptic plasticity. It also plays a role in the DNA damage response by assembling with other deacetylases to regulate histone acetylation and facilitate DNA repair.
