Zinc finger CCHC domain-containing protein 3 (ZCCHC3) is a human protein encoded by the ZCCHC3 gene, characterized by its CCHC-type zinc finger motifs. These motifs enable ZCCHC3 to bind both DNA and RNA, facilitating its role in the innate immune system's response to viral infections. Functionally, ZCCHC3 acts as a co-sensor alongside key pattern recognition receptors such as cyclic GMP-AMP synthase (cGAS), retinoic acid-inducible gene I (RIG-I), and melanoma differentiation-associated protein 5 (MDA5). Through these interactions, ZCCHC3 enhances the detection of viral nucleic acids, thereby promoting the activation of type I interferon responses and bolstering antiviral immunity. Beyond its antiviral functions, ZCCHC3 has been identified as a host restriction factor that inhibits the activity of endogenous retrotransposons, such as LINE-1 elements. This suggests a broader role in maintaining genomic stability by preventing the mobilization of potentially harmful genetic elements.
