MAPK12, also known as p38γ or ERK6, is a member of the p38 mitogen-activated protein kinase (MAPK) family, which includes p38α (MAPK14), p38β (MAPK11), p38γ (MAPK12), and p38δ (MAPK13). These kinases are integral to cellular responses to various extracellular stimuli, such as pro-inflammatory cytokines and physical stress, leading to the activation of transcription factors like ATF2 and ELK1. Unlike other p38 isoforms, MAPK12 exhibits unique functions. It plays a significant role in myoblast differentiation and negatively regulates cyclin D1 in response to hypoxia in adrenal cells, suggesting its involvement in inhibiting cell proliferation while promoting differentiation . Additionally, MAPK12 participates in the regulation of UV-induced checkpoint signaling and the repair of UV-induced DNA damage, as well as G2 arrest following gamma-radiation exposure . It also regulates glucose transporter expression and glucose uptake in skeletal muscle cells. MAPK12 is a subject of interest in various research areas, including cancer biology, neurobiology, and metabolic diseases.
