| In Vitro | Pirfenidone elicits growth-inhibitory effects and reduces TGF-beta2 protein levels in human glioma cell lines.[1] In RAW264.7 cells，a murine macrophage-like cell line，pirfenidone suppressed the proinflammatory cytokine tumor necrosis factor-alpha(TNF-alpha)by a translational mechanism.[2] Pirfenidone(0～0.5 mg/ml，24h)inhibits TGF-β2-induced proliferation，migration and epithlial-mesenchymal transition of human lens epithelial cells line SRA01/04 at nontoxic concentrations.[3] Pirfenidone inhibited fibrocyte migration by pirfenidone in vitro.[4] |
| In Vivo | In the murine endotoxin shock model，pirfenidone potently inhibited the production of the proinflammatory cytokines，TNF-alpha，interferon-gamma，and interleukin-6，but enhanced the production of the anti-inflammatory cytokine，interleukin-10.[2] Pirfenidone(300 mg/kg/day; 2 weeks; orally)attenuated the fibrocyte pool size in bleomycin-treated mouse lungs via attenuation of CCL2 and CCL12 production in vivo.[4] |
