The assay uses an SPHK-specific fluorogenic substrate, NBD-sphingosine. SPHK2 phosphorylates this substrate generating a shift in its spectral properties, which can be easily quantified using a fluorescence plate reader at excitation and emission wavelengths of 550 and 584 nm, respectively. The reversible dual SPHK1/2 inhibitor SLC5111312 is included as a positive control and inhibits SPHK2 with an IC50 value of approximately 4 μM.分子式分子量
