p16-INK4A (also known as CDKN2A) is a tumor suppressor protein that plays a key role in regulating the cell cycle. It belongs to the INK4 family of cyclin-dependent kinase inhibitors and functions primarily by inhibiting CDK4 and CDK6. These kinases are essential for the phosphorylation of the retinoblastoma (Rb) protein, which is necessary for the transition from the G1 phase to the S phase of the cell cycle. By preventing this phosphorylation, p16^INK4A effectively blocks the cell cycle progression, acting as a brake to prevent uncontrolled cell division. This function is crucial for maintaining genomic stability and preventing tumor formation. Alterations or loss of p16^INK4A expression are commonly associated with various cancers, including melanoma, pancreatic cancer, and head and neck squamous cell carcinoma.
